Studies on the characteristics of polyphenol oxidase from Aralia elata flowers

@EN The flowers of Aralia elata Seemann was homogenized with phosphate buffer(pH 5.5) in a wring blender, polyphenol oxidase was precipitated between 20~80(w/v) saturation with ammonium sulfate. After the precipitation was dialized, it was
fractionated
by Sephadex G-25 coulmn. Optimum pH of the enzyme was 7.0. Among the o-dihydroxy compounds tested, theenzyme was most reactive toward chlorogenic acid, followed by pyrocatechol, caffeic acid. M-Creasol and p-cresol were poor substrates for the
nzyme. As
copper-chelating agents sodium diethyldithiocarbamate and potassium cyanide exhibited powerful inhibition for the polyphenol oxidase from the flowers of Aralia elata Seemann. Polyphenolic compound obtained from the flowers of Aralia elata Seemann
was
identified as chlorogenic acid by GC/MS.
